Carrier questions

If I have a real hig ratio like a 2.29:1, and i want to go to a 3.73:1, do I need a new carrier? I just read an ebay auction, and that is what they said for a set of 4.10's. Auction ends soon, please help!

There are two carriers for 7.5" rears. The 2 series is good for up to 3.08. Anything higher numerically requires a 3 series carrier. It's possible to use a ring gear spacer to install higher numerical gears on a 2 series carrier but some people don't recommend it. There are also soem "thick" aftermarket gears that will fit on a 2 series carrier. However, if you want to mount factory gears you should get a 3 series carrier to properly install your 3.73 gears.

Now is there a difference in rear ends and the number of splines in the axle? I see 26 and 28 advertised on ebay for a 7.5" rear axle. Without tearing down, how would I know, or would I need to know?

All H-body 7.5" rears used 26 spline axles. The 28 spline axles were introduced mid to late 80's, in the S-10's at least. When swapping axles from an S-10 you have to look for 26 spline, 2 wheel drive axles which were used up to around the mid 80's. Now, if you purchase a new carrier you can get a 28 spline unit either used or aftermarket, then you would have to use 28 spline axles to go with it. You just have to make sure that both your carrier and axles match.
